Aim: Many research results have verified that hyaluronic acid plays an important role in repair and reestablishment of cartilage. Aim: Many research results have verified that hyaluronic acid plays an important role in repair and reestablishment of cartilage. This article was designed to evaluate the preventive effect of hyaluronic acid on degenerative articular cartilage of immobilized rabbit knees and assess its efficiency. Methods: The experiment was performed at the Basic Institute of Taishan Medical University from October 2004 to May 2005. 1 Totally 24 New Zealand male rabbits with the body mass of 2.5-3.0 kg were divided randomly into control group and hyaluronic acid group with 12 rabbits in each group. 2 The rabbits of hyaluronic acid group were injected with hyaluronic acid into left knees . The rabbits of control group were treated with the same amount of saline into left knees, once a week for 5 times. The left legs were immobilized by plaster casts after the first injection, and a port in each cast was left at the knee joint for remanent injections. The rabbits were sacrificed one week after the last injection, and cartilage of left femur condyles was harvested for paraffin sections, the counterparts of right knees were harvested simultaneously as normal control. 3 Morphologic changes of haematoxylin-eosin stained sections were observed via microscope. The average gray scales of methylaniline sections were analyzed via Image-Pro Plus 6.0 system. Fluorescence intensity of collagen type II impressed by immunofluorescence staining was analyzed via laser confocal scanning microscope. Results: Totally 24 rabbits were involved in the result analysis. In hyaluronic acid group, HE staining showed that the cartilage was not thin, alignment of chondrocytes was regular, the size of cells and staining of cellular nucleus were even, tidal line was integrated, and no pannus produced. In control group, HE staining showed that the cartilage was thin, the alignment of chondrocytes was irregular, the size of cells and staining of cellular nucleus were not even, the tidal line was not integrated and pannus invaded in cartilage basilar part. The average gray scales of methylaniline sections and fluorescence intensity of collagentype II in hyaluronic acid group were higher than those in control group , and were significantly different with normal cartilage . Conclusion: Hyaluronic acid can effectively prevent degeneration of cartilage after rabbit knee joint is fixed, however it cannot entirely avoid the degenerative changes as normal.收起
