Objective To observe the effect of PI3 K inhibitor 2-morpholino-8-phenyl-4 H-chr-omen-4-oneon the proliferation and invasion of TMZ resistant human glioma cell line U251/TR and to explore its mechanism.Method add LY294002 in cell line U251/TR cells,by MTT colorimetric assaydetection method to detect cell survival,by Transwell method to detect cell invasion,by flow cytometry to detect cell cycle,by Annexin V method and TUNEL method to detect he apoptosis.By Western blot Detection to evaluate MGMT protein expression and PI3 K/Akt signal pathway proteins such as p-AKT,MMP-2,NF-κB,Bcl-2,Caspase-3,CyclinD1.Result Compared with the blank control group and DMSO group,the survival rate of LY294002 group was significantly decreasedin second days.Compared with the blank control group and the DMSO group,the number of transwell through cell of LY294002 group was significantly decreased.The cell cycle of LY294002 group was significantly blocked in G0/G1 phase,S phase was significantly reduced,and the rate of apoptosis was increased significantly.Western blotting results show that LY294002 group can effectively inhibit the expression of P-AKT,MMP-2,MGMT,NF-κB,Bcl-2 and CyclinD1,and up regulate the expression of Caspase-3.Conclusion LY294002 targeting PI3 K/Akt signaling pathway significantly inhibited U251/TR cell proliferation,invasion,and promote cell apoptosis,and its mechanism may be related to the down-regulation of expression of PI3 K/Akt signaling pathway protein and MGMT protein by LY294002.收起
