Herein, a target-induced polarity switching photoelectrochemical biosensor was constructed by integrating the exponential amplification reaction with the CRISPR/Cas12a system to achieve more accurate detection of miRNA-622. Based on the n-type semiconductor CN modified electrode to provide initial anodic photocurrent signal, target miRNA-622 triggered the exponential amplification reaction, which converted trace amounts of miRNA-622 into a large quantity of DNA1 and DNA2 molecular. The DNA1 introduced the p-type semiconductor BiOBr nanosheets binding the DNA tetrahedron modified electrode surface to convert the PEC signal into a cathodic photocurrent. In the meantime, the DNA2 as activators bound with crRNA and Cas12a to form the ternary complex and activated the trans-cutting characteristics of Cas12a, cleaving H1-loaded BiOBr and leading to the quench of the cathodic photocurrent. Benefiting from the PEC polarity switching and EXPAR assisted CRISPR-Cas12a system, the proposed sensing platform exhibited high sensitivity with an excellent detection limit of 0.92 fM . Polarity transformation of PEC biosensor possessed the superiority of effectually avoiding the issue of false positive/negative signals in the detection process as a result of PEC signal with more accurate analysis achieved, which is promising to be applied for cancer diagnosis and treatment monitoring.收起
