Objective To explore the effects of salidroside on the expression of the Sonic hedgehog -phos-phatidylinositol 3-kinase /protein kinase B -matrix metalloproteinase-2 pathway and dopamine D2 receptor in the eye tissues of guinea pigs with lens-induced myopia. Objective To explore the effects of salidroside on the expression of the Sonic hedgehog -phos-phatidylinositol 3-kinase /protein kinase B -matrix metalloproteinase-2 pathway and dopamine D2 receptor in the eye tissues of guinea pigs with lens-induced myopia. Methods A total of 18 2-week-old female healthy tricolor guinea pigs were randomly assigned into the normal control group, the lens-induced myopia group and the lens-induced myopia + salidroside group, with 6 guinea pigs in each group. Guinea pigs in the normal control group were gavaged 2 mL of normal saline daily without any other treatment. In the LIM group and the LIM + SA group, guinea pigs used bioadhesive to attach the self-made unilateral glasses frame near the orbit of the guinea pigs, so that the - 6. 00 D lens was close to and covered the right eye of the guinea pigs to prepare the myopia model. Then,guinea pigs in the LIM group were gavaged 2 mL of normal saline daily, and guinea pigs in the LIM + SA group were gavaged salidroside 100 mg * kg daily. All three groups of guinea pigs were intervened for 4 weeks. The diopter of the right eyes of the three groups of guinea pigs was measured before modeling and 4 weeks after modeling. The expression of PI3K and AKT mRNA in the retinal tissues of the three groups of guinea pigs was deteeted by real-time fluorescence quantitative polymerase chain reaction. The expressions of Shh,PI3K,AKT and D2R proteins in the retinal tissues and the expression of MMP-2 protein in the scleral tissues of guinea pigs in the three groups were detected by Western blot. Results Before modeling, there was no significant difference in the refractive power among the three groups of guinea pigs . Four weeks after modeling, the refractive power of the guinea pigs in the LIM and LIM + SA groups was significantly higher than that in the normal control group . The relative expression levels of AKT mRNA in the retinal tissues of the guinea pigs in both the LIM and LIM + SA groups were significantly higher than those in the normal control group . The relative expression levels of PI3K and AKT mRNA in the retinal tissues of the guinea pigs in the LIM + SA group were significantly lower than those in the LIM group . Compared with the normal control group,the relative expression levels of Shh,PI3K,AKT,and MMP-2 proteins in the right eye tissues of the guinea pigs in both the LIM and LIM + SA groups were significantly increased,while the relative expression level of D2R protein was significantly decreased . Compared with the LIM group, the relative expression levels of Shh, PI3 K, AKT, and MMP-2 proteins in the right eye tissues of the guinea pigs in the LIM + SA group were significantly decreased, while the relative expression level of D2R protein was significantly increased . Conclusion Salidroside can delay the progression of myopia by inhibiting the expression of Shh-PI3K/AKT-MMP-2 pathway and promoting the expression of D2R protein.收起
